Brain Smear
Method:
Squash a small amount (˜4mm2) of grey matter between 2 slides
Place slides at right angles to each other
Move top slide sideways to the far end of the lower slide
Allow to air dry
Stain using Diff Quik
5 dips in fixative
5 dips in red solution
5 dips in blue solution
Rinse in tap water
Scan
Scan slide for capillaries
Look for erythrocytes within the capillaries that are infected with Babesia organisms [these will appear as 1 or 2 blue dots]
Safety Precautions
Wear lab coat and disposable gloves
Wash hands thoroughly when finished
Cleaning Up Procedures
Dispose of all disposable equipment in clinical waste bin or sharps as appropriate
(secure in sealed plastic bag to reduce any odour)
Wipe down work area with 70% enthanol
Ensure lab is clean and tidy
