Faecal Float - Fecalizer Technique

Use of fumehood recommended

Method:

Scoop faeces into base of container
Half fill with flotation solution and twist until sample is thoroughly mixed
Push down firmly to snap basket in place
Fill with more flotation solution until a positive meniscus is obtained
Top with coverslip and let sit for 3 to 4 minutes
Carefully lift coverslip and place onto microscope slide
Scan for eggs and oocysts
(NB: Try not to get too much froth, as it is hard to see eggs with lots of air bubbles)

Flotation Solution:

Sodium Nitrate (SG 1.20)
315g NaNO3, in 900ml to 1L dH2O until a SG of 1.2 is reached

    Alternative solution:
        Saturated Salt(NaCl) - used in student pracs

Safety Precautions

Wear lab coat and disposable gloves
Wash hands thoroughly when finished

Cleaning Up Procedures

Pour sodium nitrate into appropriate chemical waste container
Dispose of all disposable equipment in clinical waste bin or sharps as appropriate
(secure in sealed plastic bag to reduce any odour
Rinse any glassware and spray with 70% ethanol before putting in wash up container
Wipe down work area with 70% ethanol
Ensure lab is clean and tidy
